Q1:3T3-L1细胞可以用FBS培养进行扩增吗?
使用胎牛血清细胞会增殖更快,可能会过度生长,密度过饱和后可能出现细胞开始分化的情况。这个细胞日常培养注意不要过饱和,80%密度及时传代。3T3-L1使用的血清是刚出生不久的新生牛血清。

Q2:3T3-L1细胞换液后大片的细胞飘起然后大面积剥落了?
培养基使用前需要37度预热好,避免细胞预冷收缩,PBS需要恢复室温的;另外有可能是培养时间过长密度太大长太满了,细胞的贴壁性变弱,不要让这株细胞长得过满乃至开始融合,在细胞密度达到80%就传代,密度太高细胞容易飘。如果漂浮了,可以收集到离心管中进行消化,消化过程不吹散细胞团,消化完毕终止的时候吹散即可,重新接种。
